I've done two before. It didn't hurt me. But in my experience it didn't increase the urgency or make my poop come out sooner, it just seemed to make the aftereffects last longer.
For those who haven't used dulcolax, this is how it works (at least for me): after 30 minutes to an hour, the urge to poop gets pretty strong. Especially if I haven't pooped in at least a day, it results in a pretty big and unavoidable shit, and if timed right can result in an accident (which is what I wanted). But the aftereffects are analogous to "dry heaves" -- even after I am more or less empty, for some hour or so afterwards I would suddenly cramp and find myself pushing despite my best efforts. Even if I didn't want another accident and had already changed into clean clothes, I would find that I had shit myself again. With two suppositories the aftereffects were even stronger.
So I haven't bothered with 2 at a time after that first experience; one is enough for me.
